No, it's confirmed in the FAQ that we'll be seeing Twilight, Applejack, Rarity, and Pinkie Pie again. RD and Fluttershy still aren't up to showing quality.

From what I can see so far, Pinkie is a monster! A bit overpowered compared to the last I've seen of her.
The game is bad.Will never live up to the hype all indie games fail.
It’s looking great! I'm glad they are taking their time to really polish the game, and it seems like they are aiming for nigh perfect once the release happens.

Anyway I haven't really been following the production of this game but I think I'll have to start, it's looking to be a pretty solid game, and the effort the creators are putting into it shows.
I'm really hoping to see Derpy and Spike in this game. Zecora would be cool too.

Fluttershy: either she does The Stare and you explode from the d'awww or she cries and a horde of bronies instantly appears and tears you apart because "That b*st*rd hurt Fluttershy!"
Rainbow Dash: Atomic Rainbooms you into oblivion or she takes you to Cloudsdale and tosses you into the Pegasus Device.
Pinkie Pie: Either she drops a massive cake on you ("Weeee! I hope you enjoy my 'pound' cake! Of course, this cake will be pounding you!") or she drags you into the basement and then... "Cupcakes!"
Rarity: turns you into her latest fashion design. "Ponywear -- by ponies, for ponies, made /from/ ponies!"
Applejack: she kicks your heart out through your back.
Twilight Sparkle: Goes into 'inferno mode' and incinerates you, compete with post-action friendship report.
And what is that book she's reading in the sample fight? "How to beat your friends into a bloody pulp"?

So awesome, can't wait to get my hands on this game.
I am kinda confused about Rarity and Pinkie, though. Twilight is obviously a teleporter/trapper hybrid, AJ looks like a fairly straightforward brawler and Fluttershy seems like she'll be a puppeteer-type, at least according to the commentary. But I just can't place Rarity and Pinkie's style. Could someone help me out there?
Still no Rainbow Dash.. The Fluttershy duel was hilarious and the rest seems like it'll be a joy to play, though. The Dracula skin for Rarity, the Surprise skin for Pinkie and the Big Mac skin for Applejack all looked great.
