Here's the full scene. Eli, a popular radio preacher, is trying to broker a land deal with oil Tycoon Daniel. The conflict over the land had set Eli and Daniel against one since Eli was a teenager. After forcing Eli to renounce God, Daniel proceeds to berate and insult Eli, explaining that like a very long straw, he has already taken the oil from the land Eli is trying to sell. I can see why Twilight might look upset if put in a similar situation.
Now as for my own ignorance, what's the deal with Juiceboxshy?
One of the popular "ask (pony)" tumblr accounts dealt with sweet and innocent Pinkieshy shipping, and one of the earlier drawings in it featured Flittershy drinking from a juicebox. People responded very well to it, so the artist drew it some more and it soon became a meme within the ask account, then it was referenced in other tumblr accounts, then it spread to ponibooru and then ponychan and the rest is history.
