Physically? The only adult is Celestia, with Luna being in her early pubescence. Nightmare Moon reached a full physical development due the the effects of Discord, and possibly Discord himself if he has a physical body.
All the regular ponies are permanently locked in a magical 'prepubescent' state, though still able to reproduce sexually. Think Santa's elves as portrayed by child actors; the elderly ponies are akin to geriatric children, the effect being similar to that of a human childhood disease here on Earth called Progeria.
It seems adult physical development in ponies is tied to outright godhood, so the purpose of the lock on regular ponykind may have been intended as a way to end a pony-god war and/or to create a peaceful civilization ruled by a loving mother figure.
